This technique (detta anche Quickchange) is based on the use of the restriction enzyme dpn1 which è specifico per il DNA metilato di origine batterica e quindi degrada solo il plasmide parenterale e non quello neosintetizzato.

The principle steps of this method are:
  1. Denaturazione per separare i due filamenti di DNA del plasmide da mutare
  2. Riappaiamento con due oligonucleotidi mutagenici complementari (ciascuno dei quali contenente la mutazione desiderata nella stessa posizione) che faranno da primers per la successiva PCR. Il primer deve essere abbastanza lungo da permettere l'ibridazione del DNA non perfetta.
  3. PCR con la polimerasi Pfu ad alta fedeltà per ottenere due nuove molecole di DNA contenenti la mutazione, che si appaieranno per dare il plasmide mutato.
  4. Affinchè il nuovo filamento del plasmide possa saldarsi è necessario aggiungere la DNA ligasi.
  5. Digestione con l'enzima di restrizione Dpn1 in modo da degradare il DNA parentale ma da lasciare intatto quello neosintetizzato, che non è metilato poichè in vitro non vi è la DNA metilasi.
  6. Il plasmide mutagenizzato viene infine usato per trasformare delle cellule batteriche.
