Mutagenesi USE

  • Elimina il sito di restrizione unico
  • Primer Usa a causa: il primo Viene detto primer mutagenico, in quanto è il responsabile dell'introduzione della Mutazione, il secondo elimina il sito unico nel plasmide
  • Heteroduplex: lo stampo ha il sito di restrizione, mentre il filamento nuovo ha la Mutazione, ma non il sito di restrizione
  • Incubazione con enzima. si ottiene la linearizzazione di peso, ma non dell'heteroduplex
  • Trasformazione in ceppo di E. Coli che non effettua la riparazione (BMH71-18 DNA muts lineare Trasforma 10-1000 volte di più Rispetto al circolare) si ottiene la replicazione dell'heteroduplex
  • Seconda digestione e trasformazione nel ceppo di E. Coli normale (mutati 5-50%)
