  • Elimina sito di restrizione unico.
  •  Utilizzo di due Primers , il primo viene detto primer mutagenico  ed è responsabile dell'introduzione della mutazione, il secondo elimina il sito unico nel plasmide.
  • Heteroduplex: lo stampo ha il sito di restrizione, mentre il filamento nuovo ha la mutazione, ma non il sito di restrizione.
  • Incubazione con l'enzima , si ottiene la linearizzazione di wt, ma non dell'eteroduplex.
  • Trasformazione in ceppo di E. Coli che non effettua la riparazione (BMH71-18 DNA mutS il DNA linere trasforma 10-1000 volte meno Rispetto al circolare). si replica l'eteroduplex.
  • Seconda digestione e trasformazione nel ceppo di E. Coli normale (5-50% mutati).
