Richiede che il frammento di DNA in cui inserire la mutazione sia clonato in un plasmide e fiancheggiato da due siti di restrizione.Si usano due enzimi per dare una direzione precisa al filamento inserito. Taglio del plasmide con i due enzimi di restrizione.Separazione del frammento e sostituzione con una CASSETTA OLGONUCLEOTIDICA a doppio filamento che contenga la mutazione da introdurre. E' un sistema semplice, efficiente e con cui si possono ottenere mutanti multipli.L'unico problema è quello di avere siti unici di restrizione. ? ?
